SL-1-30

SL-1-30 is a second-generation flexible heteroarotinoid (Flex-Het) analog and the purified S-enantiomer of the racemic mixture SL-1-09. Derived from the first-generation Flex-Het SHetA2, SL-1-30 was designed to overcome SHetA2's limitations, such as high lipophilicity and potential liver toxicity. SL-1-30 exhibits potent anti-cancer activity against both estrogen receptor-positive (ERα+) and estrogen receptor-negative (ERα-) breast cancer cells, with particularly enhanced growth inhibitory effects in HER2-expressing cell lines. It acts by blocking cell cycle progression, specifically reducing the percentage of cells in the S-phase, which is associated with the down-regulation of key cell cycle regulatory proteins including cyclin A, cyclin B1, cyclin D1, cyclin E, and CDK2.
