Overview

SL-115154 is a preclinical-stage recombinant fusion protein developed by Shattuck Labs as part of its Antigen Receptor Chimera (ARC) platform. The molecule is a dual-sided fusion protein consisting of the extracellular domain of the Colony Stimulating Factor 1 Receptor (CSF1R) and the CD40 ligand (CD40L), linked by an Fc domain. SL-115154 is designed to modulate the tumor microenvironment by simultaneously inhibiting the immunosuppressive CSF1/CSF1R axis and activating the CD40 costimulatory pathway. By acting as a decoy receptor for CSF1, it neutralizes the recruitment and polarization of tumor-associated macrophages, while the CD40L component provides agonistic signaling to CD40 on antigen-presenting cells to enhance T-cell mediated anti-tumor immunity. The asset was originally part of a 2017 research and development collaboration with Takeda, but Shattuck regained full worldwide rights following the termination of the agreement in 2021.
