SL-172154 is an investigational Agonist Redirected Checkpoint (ARC) fusion protein designed to simultaneously inhibit the CD47/SIRPα immune checkpoint and activate the CD40 costimulatory receptor, thereby enhancing antitumor immune responses. The drug consists of a hexameric fusion protein that joins the extracellular domain of SIRPα to the extracellular domain of CD40L via an inert IgG4-derived Fc domain. By blocking the "don't eat me" signal mediated by CD47/SIRPα and stimulating immune activation through CD40, SL-172154 aims to promote both innate and adaptive anti-tumor immunity. It is being developed primarily for advanced cancers, including acute myeloid leukemia (AML), higher-risk myelodysplastic syndrome (HR-MDS), and platinum-resistant ovarian cancer. Preclinical studies have shown superior antitumor activity compared to targeting either pathway alone[1][5][6][7][8].
