SL-172154 is a first-in-class bifunctional fusion protein designed to simultaneously inhibit the immunosuppressive TGF-β pathway and activate the OX40 costimulatory pathway. It consists of the extracellular domain of the Type II TGF-β receptor (TGFβRII) fused to the OX40 ligand (OX40L) via a central Fc domain. By sequestering TGF-β1 and TGF-β3, it prevents TGF-β-mediated immune evasion and fibrosis in the tumor microenvironment. Simultaneously, the OX40L component binds to OX40 on T cells and natural killer (NK) cells, promoting their activation, proliferation, and survival. This dual mechanism aims to convert immunosuppressive "cold" tumors into immune-active "hot" tumors. It is being developed by Shattuck Labs for the treatment of various solid tumors and hematologic malignancies.
