Overview

SL-325 is a first-in-class high-affinity antagonist antibody targeting the Death Receptor 3 (DR3), a member of the tumor necrosis factor receptor superfamily. It is being developed by Shattuck Labs for the treatment of inflammatory bowel diseases (IBD), including Crohn's disease and ulcerative colitis. The drug works by blocking DR3, which is constitutively expressed on multiple immune cells that drive inflammation in IBD, thereby inhibiting the TL1A/DR3 signaling axis involved in promoting inflammation. Preclinical studies have demonstrated superior inhibition and efficacy compared to therapies targeting TL1A, with no evidence of toxicity or residual agonism observed in non-human primate toxicology studies. SL-325 shows full and durable receptor occupancy at low doses with potential for extended dosing intervals.
