SL-425 is an extended half-life monoclonal antibody targeting Death Receptor 3 (DR3, also known as Tumor necrosis factor receptor superfamily member 25). It is being developed by Shattuck Labs for the treatment of inflammatory bowel diseases (IBD). Mechanistically, SL-425 functions as a DR3 antagonist/modulator, aiming to block the DR3/TL1A pathway, which is clinically validated as a driver of inflammation in autoimmune conditions such as IBD. This second-generation molecule is an extension of Shattuck's DR3 pipeline, engineered for an extended half-life to enhance clinical utility and dosing convenience. As of May 2025, it remains in preclinical development.
