SL-701 is a multi-antigen peptide-based immunotherapy developed by Stemline Therapeutics (a subsidiary of Menarini Group) for the treatment of advanced gliomas, including glioblastoma multiforme (GBM). The vaccine consists of three synthetic short peptides corresponding to specific epitopes of tumor-associated antigens (TAAs) frequently overexpressed in malignant gliomas: interleukin-13 receptor alpha-2 (IL-13Rα2), ephrin-A2 (EphA2), and survivin. By presenting these antigens to the immune system, SL-701 aims to activate a cytotoxic T-lymphocyte (CTL) response to recognize and eliminate cancer cells. In clinical settings, it is typically administered subcutaneously alongside immune adjuvants such as GM-CSF, imiquimod, or the TLR3 agonist poly-ICLC (Hiltonol) to enhance the magnitude and durability of the anti-tumor immune response. It has received Orphan Drug Designation from the FDA for the treatment of glioma.
