SL-846 is a potent and selective small molecule inhibitor of **dual-specificity tyrosine-phosphorylation-regulated kinase 1A (DYRK1A)**. Originally developed by **Sosei Heptares** (formerly Sosei), the compound was identified as a lead candidate for the treatment of cognitive impairment associated with **Down syndrome** and **Alzheimer's disease**. DYRK1A is a key kinase encoded by a gene located on chromosome 21, and its overexpression is strongly linked to the cognitive deficits and early-onset neurodegeneration observed in individuals with Down syndrome. SL-846 works by inhibiting the kinase activity of DYRK1A, thereby modulating the phosphorylation of downstream targets such as tau protein and amyloid precursor protein (APP), which are critical to synaptic plasticity and neuronal health. Preclinical studies in mouse models of Down syndrome have demonstrated that SL-846 is brain-penetrant and capable of rescuing spatial learning and memory deficits.
