SLAMF3 inhibitors are a class of therapeutic agents designed to block the activity of Signaling Lymphocyte Activation Molecule Family member 3 (SLAMF3), also known as CD229 or Ly9. SLAMF3 is a cell-surface glycoprotein and self-ligand expressed on various hematopoietic cells, including T and B lymphocytes, where it modulates immune cell activation and adhesion. In the context of diabetic kidney disease (DKD), research has identified SLAMF3 as an insulin resistance-responsive molecule on CD4+ T cells. Its upregulation promotes T cell activation and the secretion of pro-inflammatory cytokines like IL-17, which subsequently triggers podocyte injury through the IL-17RA-PI3K-NF-κB signaling pathway. Pharmacological inhibition or genetic deletion of SLAMF3 aims to disrupt this immunometabolic axis, potentially offering a novel strategy to prevent renal structural damage and lipid accumulation in patients with diabetes.
