SLATE v2 + ipilimumab + nivolumab is a combination immunotherapy regimen under investigation for cancer treatment. It consists of: - **SLATE v2**: A personalized or off-the-shelf neoantigen mRNA vaccine designed to stimulate the immune system to recognize tumor-specific mutations. - **Ipilimumab**: A monoclonal antibody that blocks cytotoxic T lymphocyte-associated protein 4 (CTLA-4), thereby enhancing T-cell activation and proliferation. - **Nivolumab**: A monoclonal antibody that inhibits programmed cell death protein 1 (PD-1), preventing tumor cells from evading immune detection. The combination aims to synergistically enhance anti-tumor immunity by both priming the immune system against specific tumor antigens (via SLATE v2) and releasing inhibitory checkpoints on T-cells (via ipilimumab and nivolumab). Ipilimumab and nivolumab are approved checkpoint inhibitors for several cancers including melanoma, renal cell carcinoma, colorectal cancer, and non-small cell lung cancer. The addition of a neoantigen vaccine like SLATE v2 is intended to further boost the specificity and potency of the anti-tumor response.
