SLC-391 is an orally bioavailable and highly selective small molecule inhibitor of the receptor tyrosine kinase AXL (also known as UFO), developed for its potential immunostimulating and antineoplastic activities. By binding to and preventing activation of AXL, SLC‑391 blocks downstream signal transduction pathways involved in tumor cell growth, proliferation, migration, and immunosuppression. AXL is overexpressed in many tumor types and immune cells such as macrophages, NK cells, and regulatory T-cells; its expression is associated with drug resistance and poor prognosis. Preclinical studies show that SLC‑391 inhibits AXL/PI3K/AKT-dependent cell proliferation/survival. Combination with PD‑1 inhibitors has demonstrated synergistic anti-tumor effects in models. Clinical trials are ongoing for advanced solid tumors including non-small cell lung cancer.
