OMass Therapeutics is developing a small molecule inhibitor of SLC15A4 (Solute Carrier Family 15 Member 4) for the treatment of systemic lupus erythematosus (SLE) and other interferonopathies (IFN-opathies). SLC15A4 is an endolysosomal transporter primarily expressed in plasmacytoid dendritic cells (pDCs) and B cells, where it is essential for the signaling of endosomal Toll-like receptors (TLR7 and TLR9). By transporting histidine and other ligands, SLC15A4 facilitates the recruitment of the TASL adapter protein and the subsequent activation of IRF5, leading to the production of Type I interferons and pro-inflammatory cytokines. The program utilizes OMass's proprietary native mass spectrometry platform (OdyssION™) to identify small molecules that can disrupt the function of this membrane protein, aiming to suppress the autoimmune response driven by interferon overproduction. It is currently in the discovery and preclinical development stages.
