Sleeping beauty transposed pbl is an investigational cell therapy that utilizes the Sleeping Beauty (SB) transposon system to genetically engineer autologous peripheral blood lymphocytes (PBLs), typically T cells, for cancer immunotherapy. The SB system is a synthetic DNA transposon platform designed to introduce specific DNA sequences into the genome of vertebrate cells via a "cut-and-paste" mechanism mediated by the SB transposase enzyme. In this context, patient-derived T cells are engineered ex vivo using the SB system to express tumor-specific receptors—such as chimeric antigen receptors (CARs) or T-cell receptors (TCRs)—targeting mutated neoantigens in cancer. These modified T cells are then expanded and reinfused into patients with metastatic cancers, including small cell neuroendocrine carcinoma of the bladder, breast carcinoma, and cervical carcinoma[1][2][3][5]. The approach aims to enhance anti-tumor immune responses by redirecting patient immune cells against malignant targets. Developed by the National Cancer Institute, this therapy has been investigated in clinical trials for various metastatic cancers.
