Sleeping Beauty transposed peripheral blood lymphocytes are autologous or allogeneic T lymphocytes genetically modified ex vivo using the Sleeping Beauty (SB) DNA transposon system. The SB system is a non-viral gene transfer platform that uses a transposase enzyme to stably integrate therapeutic genes, such as chimeric antigen receptors (CARs) or specific T-cell receptors (TCRs), into the genome of peripheral blood lymphocytes. The modified lymphocytes are expanded ex vivo and reinfused into patients for adoptive immunotherapy, primarily for the treatment of various cancers, especially hematological malignancies and some solid tumors. Mechanistically, the SB transposon system combines the advantages of viral and naked DNA vectors, allowing stable and efficient genetic modification without the risks associated with viral vector production or integration site bias. The approach enables sustained transgene expression and is under investigation as a platform for cell-based cancer immunotherapy, notably CD19 CAR T-cell therapy[1][2][3][4][5].
