SLIT2 is a recombinant version of the endogenous Slit guidance ligand 2 protein, which serves as a ligand for the Roundabout (ROBO) family of receptors. Traditionally known for its role in axon guidance during neural development, SLIT2 has been identified as a tumor suppressor in various hematological malignancies, including acute myeloid leukemia (AML). In AML, the SLIT2 promoter is frequently hypermethylated, leading to reduced expression and poor prognosis. Recombinant SLIT2 protein therapy aims to restore this signaling axis, resulting in inhibited cell proliferation, induction of G1 phase cell cycle arrest, and reduced colony formation. Preclinical studies have demonstrated that systemic administration of recombinant SLIT2 can delay leukemogenesis, reduce leukemic infiltration in the bone marrow and spleen, and improve overall survival in vivo.
