SLN226 is an investigational small interfering RNA (siRNA) therapeutic developed by Silence Therapeutics (formerly Silencio) designed to target and silence the expression of complement component 3 (C3). Utilizing the company's proprietary mRNAi GOLD (GalNAc Oligonucleotide Discovery) platform, SLN226 is engineered to specifically inhibit the production of C3 protein in the liver. By degrading C3 mRNA before it can be translated, the drug aims to block the complement cascade at its central convergence point, providing a potential treatment for various complement-mediated disorders such as C3 glomerulopathy. The program has been part of a strategic collaboration with Mallinckrodt Pharmaceuticals. While SLN226 was an earlier designation in the pipeline, Silence Therapeutics has also advanced SLN501, an optimized version of the C3-targeting siRNA candidate.
