Overview

SLOG is a multi-agent chemotherapy regimen consisting of S-1 (a combination of tegafur, gimeracil, and oteracil), leucovorin (folinic acid), oxaliplatin, and gemcitabine. It is primarily developed and investigated by the Taiwan Cooperative Oncology Group (TCOG) and the National Health Research Institutes (NHRI) for the treatment of advanced gastrointestinal malignancies, particularly pancreatic ductal adenocarcinoma (PDAC) and biliary tract cancers. The regimen is designed to provide a potent combination of antimetabolites and platinum-based agents, often compared to modified FOLFIRINOX (mFOLFIRINOX) in clinical trials to evaluate its efficacy and safety profile as a first-line treatment option. Recent studies have also explored SLOG as a backbone for combination therapies with allogeneic NK cells.
