SLS009 + venetoclax + azacitidine is a **triple combination regimen** under investigation for treatment of relapsed/refractory acute myeloid leukemia (AML). **SLS009 (tambiciclib)** is a highly selective **CDK9 inhibitor** with improved safety and potency designed to disrupt transcriptional regulation in cancer cells, including those with high-risk molecular features. **Venetoclax** is a selective **BCL-2 inhibitor** that induces apoptosis in AML cells. **Azacitidine** is a **nucleoside metabolic inhibitor** and a DNA methyltransferase inhibitor used to induce cell death by DNA hypomethylation and cytotoxicity. This combination is evaluated for patients with AML, including those with myelodysplasia-related changes or resistance to venetoclax-based regimens. The drugs are provided intravenously or subcutaneously in a clinical setting.
