Overview

SLTE-1009 (also known as DS009) is a clinical-stage monoclonal antibody being developed for the preventative treatment of migraine and other headache disorders. Originally developed by DartsBio Pharmaceuticals and in-licensed by Slate Medicines (which merged with Fulcrum Therapeutics in August 2026), SLTE-1009 targets both pituitary adenylate cyclase-activating polypeptide (PACAP) and vasoactive intestinal peptide (VIP). This dual-targeting mechanism distinguishes it from currently approved migraine therapies that primarily target the calcitonin gene-related peptide (CGRP) pathway. The antibody is engineered with half-life extension technology to enable convenient, infrequent subcutaneous dosing, potentially allowing for at-home administration. It has received clearance for Phase 1 clinical trials in Australia.
