SLX-3065 is an orally bioavailable, potent, and selective molecular glue degrader of Cyclin-Dependent Kinase 9 (CDK9), developed by Biolexis Therapeutics. As a molecular glue, it facilitates the interaction between CDK9 and an E3 ubiquitin ligase, leading to the ubiquitination and subsequent proteasomal degradation of the kinase. CDK9 is a critical component of the positive transcription elongation factor b (P-TEFb) complex, which regulates the transcription of short-lived oncogenic proteins such as MYC and MCL-1. By degrading CDK9, SLX-3065 aims to treat aggressive variant prostate cancers (AVPC) and other MYC-driven malignancies. The compound was featured in a late-breaking research poster at the 2024 American Association for Cancer Research (AACR) Annual Meeting.
