SLX-9029 is a novel, selective small molecule inhibitor of Transglutaminase 2 (TGM2) being developed by Biolexis Therapeutics for the treatment of glioblastoma. TGM2 is a calcium-dependent enzyme that facilitates post-translational modifications by cross-linking extracellular matrix (ECM) proteins, thereby stabilizing the tumor microenvironment and contributing to therapeutic resistance. SLX-9029 was identified through Fragment2Lead (F2L) optimization using the MolecuLern fragment library. In preclinical studies, it demonstrated an IC50 of 1.2 µM in cell-free assays and showed efficacy in U138 and U87 glioblastoma cell lines. The compound aims to modulate the tumor microenvironment and enhance immune responses against glioblastoma.
