SLX-9031 is a novel, selective small molecule inhibitor of Transglutaminase 2 (TGM2) being developed by Biolexis Therapeutics for the treatment of glioblastoma. TGM2 is a calcium-dependent enzyme that cross-links extracellular matrix (ECM) proteins, contributing to tumor microenvironment stability, cell proliferation, and resistance to therapy. SLX-9031 was identified via fragment-based lead optimization from the MolecuLern library and has shown an IC50 of 1.5 uM in cell-free assays, along with promising activity in U138 and U87 glioblastoma cell lines. The compound is currently undergoing preclinical evaluation, including ADME and in vivo pharmacokinetic studies, to assess its potential as a targeted therapy for glioblastoma.
