Overview

SM-88 (racemetyrosine) is an investigational oral small molecule amino acid analogue developed by Tyme, Inc. (now part of Syros Pharmaceuticals). It is designed to selectively target cancer cells by exploiting the Warburg effect. SM-88 is a non-functional isomer of tyrosine that cancer cells selectively uptake; once internalized, it interferes with protein synthesis pathways and the production of the protective transmembrane protein Mucin 1 (MUC1). This disruption increases intracellular oxidative stress and leads to apoptosis. In clinical trials, SM-88 is typically administered as part of a combination regimen known as MPS, which includes low doses of methoxsalen, phenytoin, and sirolimus to enhance its therapeutic activity. The drug has been evaluated in various advanced solid tumors, including biochemically recurrent prostate cancer and advanced pancreatic cancer.
