Sm-p80 is the large (80 kDa) subunit of a calcium-dependent neutral protease (calpain) expressed by *Schistosoma mansoni*. It is a leading vaccine candidate for schistosomiasis and has been evaluated in multiple preclinical models including mice, hamsters, and baboons. The protein plays a critical role in parasite membrane biogenesis and immune evasion. As an antigenic target for vaccination, recombinant Sm-p80 formulated with adjuvants such as glucopyranosyl lipid A (GLA-SE or GLA-Alum) induces robust humoral responses and provides significant prophylactic efficacy by reducing worm burden and egg output after challenge with *S. mansoni*. The vaccine also demonstrates cross-species protection against other schistosome species (*S. haematobium*, *S. japonicum*). Mechanistically, it elicits mixed Th1/Th2/Th17 immune responses via TLR4 or TLR9 agonist adjuvants[1][2][3][5][7].
