Sm-TSP-2 is a recombinant protein vaccine candidate designed to prevent intestinal schistosomiasis caused by *Schistosoma mansoni*. The vaccine targets a 9 kDa recombinant protein corresponding to the large extracellular loop (EC2) of the tetraspanin-2 protein, which is found on the tegument (outer surface) of the parasite and on its secreted extracellular vesicles. Sm-TSP-2 plays a critical role in parasite membrane integrity and host-parasite communication. The vaccine works by eliciting high titers of antigen-specific IgG antibodies (particularly IgG1 and IgG3) that interfere with the parasite's protective surface and block the internalization of parasite-derived vesicles by host immune and vascular cells. This immune response results in a significant reduction in adult worm burden and egg production. Sm-TSP-2 is typically formulated with Alhydrogel and sometimes supplemented with adjuvants such as the TLR4 agonists AP 10-701 or GLA-AF to enhance potency and durability of the immune response.
