The Sm-TSP-2 vaccine is an adjuvant-formulated recombinant protein subunit vaccine developed by the Access to Advanced Health Institute (AAHI), formerly the Infectious Disease Research Institute (IDRI), in collaboration with the Sabin Vaccine Institute and Baylor College of Medicine. The vaccine is designed to provide protective immunity against *Schistosoma mansoni*, a major cause of intestinal schistosomiasis. It targets the Sm-TSP-2 (tetraspanin-2) protein, which is highly expressed on the tegument (outer surface) of the adult parasite and is essential for its structural integrity and nutrient uptake. By inducing specific IgG antibodies against Sm-TSP-2, the vaccine aims to disrupt the parasite's surface membrane, leading to a reduction in worm burden and a decrease in the production of eggs, which are responsible for the inflammatory pathology of the disease. The formulation typically includes AAHI's proprietary adjuvant GLA-SE, a synthetic Toll-like receptor 4 (TLR4) agonist in a stable oil-in-water emulsion, to enhance the magnitude and durability of the immune response.
