SM1500 is an orally bioavailable, potent, and selective small molecule inhibitor of Ubiquitin-Specific Protease 1 (USP1), developed by SIMR Biotechnology for the treatment of advanced solid tumors. USP1 is a deubiquitinating enzyme that plays a critical role in the DNA damage response (DDR) pathway by regulating the stability of proteins involved in Fanconi Anemia (FA) and Translesion Synthesis (TLS). By inhibiting USP1, SM1500 promotes the accumulation of ubiquitinated substrates, leading to genomic instability and inducing synthetic lethality in cancer cells, particularly those with homologous recombination deficiency (HRD) or BRCA mutations. The drug has received Fast Track Designation from the FDA and is currently being evaluated in Phase 1 clinical trials to determine its safety, tolerability, and preliminary efficacy in patients with advanced malignancies.
