SM15685 is a highly selective, potent, orally bioavailable, and brain-penetrant small-molecule inhibitor of dual-specificity tyrosine-phosphorylation-regulated kinase 1A (DYRK1A) and 1B (DYRK1B). Originally developed by Biosplice Therapeutics and subsequently advanced by its subsidiary TenaRx, SM15685 is designed to target the underlying pathology of Alzheimer's disease and other tauopathies. By inhibiting DYRK1A/B, the drug reduces the hyperphosphorylation of Tau protein, thereby preventing the formation of neurofibrillary tangles and tau aggregates. Preclinical studies in transgenic mouse models have demonstrated that SM15685 significantly reduces tau pathology and neurodegeneration, supporting its advancement toward IND-enabling studies.
