SM17 is a novel, first-in-class humanized IgG4-kappa monoclonal antibody that targets the interleukin-17 receptor B (IL-17RB), also known as the interleukin-25 (IL-25) receptor[4][5][6][7]. By binding to IL-17RB, SM17 blocks signaling from IL-25, an "alarmin" cytokine that drives type 2 T helper cell (Th2) inflammation implicated in diseases such as asthma and atopic dermatitis[1][4][5]. This blockade suppresses Th2 immune responses by reducing the release of cytokines like IL-4, IL-5, IL-9, and IL-13 and inhibiting eosinophilia and inflammatory cell infiltration in affected tissues[1][5]. Preclinical studies demonstrated efficacy in models of allergic asthma and atopic dermatitis. In Phase I clinical trials in healthy volunteers, SM17 showed a favorable safety profile with no drug-related serious adverse events reported[1][3][5]. The drug is being developed primarily for severe asthma but is also under investigation for idiopathic pulmonary fibrosis (IPF), atopic dermatitis (AD), and other immunological disorders[4].
