Overview

SM934 is a water-soluble derivative of artemisinin developed as an orally active small molecule immunosuppressant. It exhibits potent antiproliferative and anti-inflammatory properties in vitro and in vivo. Mechanistically, SM934 acts as an antagonist of Toll-like receptor 7 (TLR7) and Toll-like receptor 9 (TLR9), modulating immune responses by inhibiting T cell activation, proliferation, and cytokine production. Preclinical studies have demonstrated its ability to suppress T cell proliferation, inhibit interferon-gamma production, promote apoptosis in activated CD4+ T cells, and block Akt phosphorylation downstream of interleukin-2 signaling. These effects contribute to its immunosuppressive activity relevant for autoimmune diseases such as systemic lupus erythematosus (SLE). Clinical development has reached Phase 2 for SLE.
