Overview

Smad6 shRNA is a short hairpin RNA (shRNA) therapeutic candidate designed to silence the expression of the SMAD6 gene. Smad6 is an inhibitory member of the Smad family that negatively regulates Bone Morphogenetic Protein (BMP) and Transforming Growth Factor-beta (TGF-β) signaling pathways. In pancreatic cancer, particularly in cases where SMAD4 is wild-type, Smad6 is frequently upregulated and acts as a pro-oncogenic factor by promoting cell proliferation, migration, and invasion. By utilizing RNA interference (RNAi) to reduce Smad6 protein levels, this shRNA construct aims to restore BMP signaling responsiveness and inhibit the metastatic progression of pancreatic adenocarcinoma. Research into Smad6 shRNA has primarily been conducted in preclinical models to validate Smad6 as a viable therapeutic target in SMAD4-intact pancreatic cancers.
