Drug intelligence / Profile preview
small EV-PH20 (also known as Exo-PH20) is a preclinical-stage engineered exosome (small extracellular vesicle) displaying GPI-anchored PH20 hyaluronidase on its surface. Developed by ShiftBio and originally researched at the Korea Institute of Science and Technology (KIST), the therapeutic candidate is designed to degrade high-molecular-weight hyaluronan (HA) within the tumor extracellular matrix. By breaking down this physical barrier, small EV-PH20 enhances the penetration of therapeutic agents and immune cells into the tumor microenvironment. Additionally, the resulting low-molecular-weight hyaluronan fragments stimulate the maturation and migration of CD103+ dendritic cells, boosting tumor-specific CD8+ T-cell responses. In preclinical studies, small EV-PH20 has been evaluated in combination with PD-L1 blockade to synergistically enhance antitumor immunity and inhibit tumor growth in cancer models.
