Overview

small interfering RNA (KDM4B) is an experimental RNA interference (RNAi) agent designed to silence the expression of the histone lysine demethylase 4B (KDM4B). KDM4B is an epigenetic co-activator of estrogen receptor alpha (ERα) signaling and is frequently overexpressed in breast cancer, contributing to tumor progression and endocrine resistance. By targeting KDM4B mRNA for degradation, this siRNA restores repressive H3K9me3 histone marks, downregulates ERα expression, and induces G1/S cell-cycle arrest. Research indicates that KDM4B knockdown also attenuates glycolytic activity, suggesting a role in metabolic reprogramming. It is primarily being investigated as a therapeutic strategy to overcome resistance to endocrine therapies like tamoxifen in ERα-positive breast cancer.
