Small interfering RNA targeting Mic60 is an experimental RNA interference (RNAi) agent designed to silence the expression of Mic60, also known as Mitofilin or IMMT. Mic60 is a critical structural component of the Mitochondrial Contact Site and Cristae Organizing System (MICOS) complex, which maintains mitochondrial cristae architecture and respiratory function. In breast cancer research, this siRNA is used as a molecular tool to investigate the consequences of Mic60 loss, which is associated with aggressive disease. Silencing Mic60 has been shown to induce the expression of epithelial-mesenchymal transition (EMT) master regulators (such as Twist, Snail, and Zeb2) and activate the NF-κB signaling pathway. This molecular reprogramming leads to increased tumor cell migration and invasion in vitro and accelerated tumor growth in vivo. While primarily used to study mitochondrial metabolic reprogramming and tumor progression, the downmodulation of Mic60 appears to promote rather than inhibit cancer aggressiveness.
