Small interfering RNA (siRNA) targeting PIEZO1 is a molecular tool and potential therapeutic agent designed to silence the expression of the PIEZO1 gene through the RNA interference (RNAi) pathway. PIEZO1 is a mechanosensitive ion channel that plays a pivotal role in sensing mechanical forces and translating them into biochemical signals, particularly in pulmonary artery smooth muscle cells (PASMCs). Research indicates that PIEZO1 regulates smooth muscle cell plasticity and glycolytic metabolism, contributing to the phenotypic switching observed in conditions like pulmonary hypertension and pulmonary fibrosis. By specifically degrading PIEZO1 mRNA, this siRNA reduces the protein levels of the channel, thereby modulating the cellular response to mechanical stretch and potentially halting disease progression.
