Small interfering RNA targeting PRDM1 (si-PRDM1) is an experimental RNA interference (RNAi) agent designed to silence the expression of the PR domain-containing protein 1 (PRDM1) gene. PRDM1, also known as BLIMP1, is a zinc finger protein that acts as a transcriptional regulator and has been implicated in the progression of various malignancies. In the context of renal cell carcinoma (RCC), PRDM1 is frequently upregulated and contributes to tumor growth and drug resistance. The mechanism of si-PRDM1 involves the sequence-specific degradation of PRDM1 mRNA, leading to reduced protein levels. This knockdown has been shown to induce ferroptosis and enhance the sensitivity of RCC cells to the tyrosine kinase inhibitor sunitinib by inhibiting the transcription of ESM1 and subsequently modulating the PI3K/Akt signaling pathway. Preclinical studies in xenograft animal models have demonstrated that PRDM1 silencing can exert significant anti-tumor effects.
