Overview

Small interfering RNA targeting USP14 is an RNA-based therapeutic designed to silence the expression of the USP14 gene. USP14 is a major deubiquitinating enzyme (DUB) associated with the 26S proteasome, where it regulates protein degradation by removing ubiquitin chains from substrates before they enter the proteasome. Overexpression of USP14 has been implicated in the progression and chemoresistance of various cancers, including non-small cell lung cancer (NSCLC). By reducing USP14 levels, this siRNA promotes the degradation of oncogenic proteins and sensitizes tumor cells to chemotherapeutic agents like cisplatin. Research has explored specialized delivery systems, such as aptamer-guided nano-zinc carriers, to improve the stability and tumor-specific uptake of the siRNA in drug-resistant models.
