Small interfering RNA targeting YB-1 is an RNA-based therapeutic approach designed to silence the expression of Y-box binding protein-1 (YB-1), a multifunctional protein that is highly expressed in various human malignancies, including glioblastoma (GBM). YB-1 acts as a key transcription and translation factor that regulates the expression of genes involved in cell proliferation, survival, and the maintenance of cancer stem cell (CSC) populations. By utilizing the RNA interference (RNAi) pathway, these siRNAs trigger the degradation of YB-1 mRNA, leading to a reduction in YB-1 protein levels. This inhibition has been shown in preclinical studies to attenuate the expression of neural stem cell markers (such as Sox-2, nestin, and musashi), reduce neurosphere growth, and induce differentiation in brain cancer stem cells, thereby potentially limiting tumor recurrence and progression.
