The smallpox alphavaccine is an investigational biodefense vaccine developed by AlphaVax using its proprietary Viral Replicon Particle (VRP) technology. The vaccine platform is derived from an attenuated Venezuelan Equine Encephalitis (VEE) virus, where the viral structural genes are replaced with sequences encoding specific smallpox (Variola virus) antigens, such as L1R, A33R, B5R, and A27L. These replication-deficient VRPs function as a delivery vehicle for replicon RNA; once they infect host cells, the RNA self-amplifies and produces high levels of the target smallpox antigens without generating new infectious virus particles. This mechanism is designed to induce a potent and balanced immune response, including both neutralizing antibodies and cellular immunity, providing a safer alternative to traditional live-vaccinia virus vaccines for protection against smallpox.
