Smallpox vaccine (Lister strain) is a second-generation, live-attenuated vaccine developed by Sanofi Pasteur for the prevention of smallpox. Derived from the historical Lister (Elstree) strain of the vaccinia virus, this version is produced using modern cell-culture technology (typically Vero cells) rather than the traditional calf-lymph method used for first-generation vaccines like Dryvax. This manufacturing process ensures a more sterile and consistent product. The vaccine is administered via scarification (percutaneous) using a bifurcated needle, inducing a 'take' (a localized lesion) that signifies successful immunization and the development of cross-protective immunity against the variola virus. It was developed primarily for the US Strategic National Stockpile as a biodefense measure.
