Smallpox Vaccine MRC-5 is a cell culture–derived live vaccinia virus smallpox vaccine produced by infecting human diploid lung fibroblasts of the MRC-5 cell line with the New York City Board of Health (NYCBH) vaccinia strain under serum-free conditions and formulated as a freeze‑dried product. It is a so‑called “second‑generation” smallpox vaccine designed as a modern replacement for calf‑lymph vaccines such as Dryvax, with improved manufacturing control and safety based on a well‑characterized human diploid cell substrate. The vaccine induces protective immunity by replicating locally after percutaneous administration and eliciting robust humoral and cellular immune responses against multiple intracellular mature virion and extracellular enveloped virion antigens of vaccinia, thereby conferring cross‑protection against variola virus, the causative agent of smallpox.[5][7][13][14]
