Overview

SMAR-T (Small Molecule Antigen-Retrieval Technology) is a therapeutic platform developed by EnGeneIC that utilizes EnGeneIC Dream Vectors (EDVs) to deliver cytotoxic payloads directly to cancer cells. The technology involves the use of bacterially derived nanocells (EDVs) that are loaded with chemotherapy drugs or other small molecules. These nanocells are then targeted to specific tumor cell surface receptors using bispecific antibodies, where one arm of the antibody binds to the EDV and the other arm binds to a tumor-specific antigen (such as EGFR). Once internalized via endocytosis, the EDV releases its payload within the intracellular environment, thereby increasing the local concentration of the drug while minimizing systemic toxicity. This approach is designed to overcome multi-drug resistance and improve the therapeutic index of conventional small molecule agents.
