SMARCA4 knockout CART19 is an experimental CD19-directed chimeric antigen receptor T cell (CART19) product in which the SMARCA4 gene, a core ATPase subunit of the SWI/SNF chromatin-remodeling complex, has been genetically deleted in the CAR T cells to enhance their antitumor function. Preclinical work has shown that SMARCA4-deficient CART19 cells display increased cytotoxicity against CD19-positive targets and higher interferon-γ production compared with conventional CART19, suggesting that SMARCA4 loss reprograms T cell chromatin to favor effector differentiation and activity.[3][7] This next‑generation “armored” CAR T approach is being investigated at the research stage as a strategy to improve outcomes of CD19 CAR T therapies for B cell malignancies.
