Overview
SMART-ADC (Selective Modulation of ADC Tumor Targeting) is an affinity-tunable antibody-drug conjugate (ADC) platform being developed by Mabswitch for the treatment of cancer, with a lead preclinical program targeting high-grade serous ovarian cancer. The SMART-ADC technology is designed to provide temporal and spatial control over target engagement. It allows the ADC to initially circulate in a lower-affinity 'OFF' state to optimize biodistribution and minimize off-target toxicity in normal tissues, followed by a ligand-triggered transition to a higher-avidity 'ON' state that promotes tumor-specific binding, internalization, and payload delivery.
