Overview

SMART CAR-T Claudin 18.2 (GC502) is an autologous chimeric antigen receptor T-cell (CAR-T) therapy developed by Gracell Biopharma (now a subsidiary of AstraZeneca) using its proprietary SMART CAR-T (Self-activating Multi-functional Anti-tumor Resistance T-cell) platform. This therapy is engineered to target Claudin 18.2 (CLDN18.2), a tight junction protein that is highly expressed in various solid tumors, including gastric, gastroesophageal junction, and pancreatic cancers, while having limited expression in normal tissues. The SMART CAR-T platform is specifically designed to overcome the immunosuppressive tumor microenvironment (TME) and enhance the persistence and expansion of CAR-T cells in solid tumors, which have historically been challenging for cell therapies. GC502 is currently being evaluated in investigator-initiated clinical trials (IIT) to assess its safety and preliminary efficacy in patients with CLDN18.2-positive advanced solid tumors.
