SMART101 is an investigational allogeneic human T lymphoid progenitor (HTLP) cell therapy developed by Smart Immune to accelerate immune reconstitution in patients following hematopoietic stem cell transplantation (HSCT). The therapy is produced using the proprietary ProTcell platform, which utilizes a 7-day ex vivo culture system featuring the Notch ligand Delta-like 4 (DLL4-Fc) to differentiate donor-derived CD34+ hematopoietic stem cells into CD7+ T-cell progenitors. Once infused into the patient, these progenitors are designed to migrate to the thymus and mature into a full, self-tolerant, and polyclonal T-cell repertoire. This approach aims to reduce the typical 12-to-18-month recovery period for the immune system to approximately 100 days, thereby potentially decreasing the incidence of life-threatening infections and disease relapse in patients with hematological malignancies such as acute myeloid leukemia (AML) and acute lymphocytic leukemia (ALL).
