**SMC021** is an investigational oral tablet formulation of recombinant salmon calcitonin formulated with 5-CNAC, an Eligen-derived gastrointestinal peptide-absorption enhancer. Salmon calcitonin is a peptide agonist of the calcitonin receptor that suppresses osteoclast-mediated bone resorption; 5-CNAC enables oral peptide delivery rather than serving as a separate therapeutic active ingredient. SMC021 was developed for postmenopausal osteoporosis and knee osteoarthritis, reached Phase III testing in both settings, and its clinical development program was subsequently shut down after failure to meet key efficacy endpoints.
