Overview
**SMNP** is an investigational nanoparticle vaccine adjuvant composed of saponin and monophosphoryl lipid A. It enhances vaccine-induced innate and adaptive immune responses; MPLA provides Toll-like receptor 4 agonism, while the particulate saponin-containing formulation promotes antigen delivery to draining lymph nodes and supports germinal-center, B-cell, and T-cell responses. SMNP originated from research led by Darrell Irvine at the Massachusetts Institute of Technology and is being evaluated with the HIV-1 envelope trimer immunogen N332-GT5 gp140 in the NIAID-sponsored phase 1 HVTN 144 study for HIV prevention.
