Overview

SMol006 is a proprietary small molecule GSPT1 (G1 to S phase transition 1) degrader developed by Orum Therapeutics. It functions as a molecular glue that binds to the Cereblon (CRBN) E3 ubiquitin ligase and drives the recruitment and subsequent proteasomal degradation of GSPT1, a translation termination factor essential for the survival of many tumor cells. SMol006 is characterized by high potency and selectivity for GSPT1. Rather than being developed as a standalone therapeutic, SMol006 is utilized as the payload component in Orum's Dual-Precision Targeted Protein Degradation (TPD) platform to create degrader-antibody conjugates (DACs). These DACs, such as ORM-5029 (targeting HER2) and ORM-6151 (targeting CD33), are designed to deliver the GSPT1 degrader specifically to cancer cells, thereby minimizing systemic toxicity while maximizing anti-tumor activity.
