SMP-534 is a novel small molecule that acts as an antifibrotic agent. It inhibits transforming growth factor-beta (TGF-beta)-induced extracellular matrix (ECM) production, particularly in fibroblast cells, and suppresses fibrosis by interfering with downstream signaling, specifically p38 mitogen-activated protein kinase activation. It dose-dependently ameliorates glomerular fibrosis, reduces urinary albumin and type IV collagen excretion, and limits mesangial expansion. SMP-534 has demonstrated efficacy in experimental animal models of chronic kidney disease and diabetic nephropathy, both alone and in combination with losartan. It is administered orally and is being researched as a therapy for chronic kidney disease and diabetic nephropathy[1][3][5][8][9].
